The FCS2 and FCS3 Chambers are closed system, parallel plate, flow-cells.  Therefore, the cells and medium are both isolated from atmospheric exposure. Cells are placed between two glass plates forming a fluid optical cavity where one of these plates is a coverslip and the other plate, called a Microaqueduct Slide, facilitates perfusion and temperature control if needed.  The perfusable optical cavity is held together by a sealing mechanism that interfaces the microscope stage.  Temperature is maintained with the aid of a two channel electronic controller. One channel controls the temperature of the metal base of the chamber.  The other controls the temperature of the Microaqueduct Slide thereby providing accurate and uniform temperature control across the entire imaging field.  These systems are optically compatible with nearly all modes of light microscopy, have near laminar flow, and user definable volume and flow characteristics making them an ideal choice for almost any imaging scenario where cells need to be perfused in an atmospherically sealed chamber. The FCS2 is compatible with inverted microscopes and the FCS3 is made for upright microscopes.

Chamber Accessories:

  • FCS2 Low Dead Volume Top (used to reduce the amount of media needed to access the optical cavity)
  • FCS2 Cooling Adapter (used to provide an additional fluid exchange for cooling purposes above and in fluid contact with the Microaqueduct Slide)
  • FCS2 Open Mode Adapter (used to allow the FCS2 base to be used like a cover glass bottomed open culture dish with wide open access through the top for micro manipulators)
  • Cooled FCS2 (CFCS2) (A modification to the base of a FCS2 where tubing is installed to facilitate the circulation of a cooled fluid to reduce the temperature of the metal base of the chamber below ambient)
  • Custom Microaqueduct Slides (can be made to facilitate flow profiles other than what is possible with the standard Microaqueduct Slide)
  • Custom gaskets (are also made to facilitate flow profiles than what is possible with the standard gasket set)
  • Extended range temperature control (temperatures above 50 degree C require specialized electronics available from Bioptechs, provide the temperature protocol and call for a quote)

 

40mm Coverslips
The disposable coverslip where you culture your cells to insert into the FCS2 or FCS3 chamber. Coverslips made from German Desag 263 low alkaline glass, they are #1.5 coverglass thick ~170 microns, and are 40mm in diameter. These coverslips come in multiple size packs:

Please specify the die number that is located on the gasket drawing below and the thickness of the gasket when ordering. Gaskets are sold 5 gaskets per pack. Microaqueduct Slides – 130119-5 – $522.45

This slide is the heart of the FCS2 and FCS3 systems. The slide has an ITO coating on the opposite side of the glass where the cells are located when assmebled, which provides intrinsic heat directly to the cells. The slide has two “T” shaped grooves cut into the inner surface of the side. The “T” groove allows the media to seek the path of least resistance and become nearly laminar before flowing across the cells. Microaqueduct slides are sold five per package.

FCS2 Chamber Flow Cell – 03060319-2-NH – $2,686.86

The FCS2 & FCS3 flow cells are so universal they can meet the needs of nearly all flow cell applications.  The flow cell is easily modified with a variety of gasket geometries (see additional gaskets above for a few samples), and a gasket thicknesses ranging from 0.1mm to 1mm. If the entire scope is in an environmental enclosure or, doesn’t need temperature control, the FCS2 is available in the following configuration and provides a substantial savings without the temperature control:

Open Mode Top for the FCS2 – 060319-2-1513 – $260.54

The open mode option allows for the FCS2 to be assembled without the microaqueduct slide thus exposing the cell on the cover slip for microinjection. The coverslip can then be removed and reassembled with the microaqueduct slide for long term, time-lapse. Click here for Instructions

Low Dead Volume Top for the FCS2

The Low Dead Volume Top is specifically designed to reduce the volume of media in the ports of the FCS2 from the point where the supply tubing attaches to the FCS2 through to the optical cavity where the cells reside for imaging. It is commonly used when working with expensive compounds and primers. Note: When using a Low Dead Volume Top, only the aperture and length of the tubes are reduced.  The rest of the FCS2 is the same as the standard model.  The following charts and examples illustrate the differences. The standard FCS2 has a minimal cross sectional area of 0.608 sq mm.  The inside diameter of the vertical tube provides ample flow for most experiments; on the order of fractional to milliliters/sec. However, the lowest dead volume top has a minimal cross sectional area of 0.196sq mm.

How it works:

Cells in the FCS2 are encased in an optical cavity bounded by the coverslip and the Microaqueduct Slide.  The flow of a coolant is facilitated above and in contact with the Microaqueduct Slide which shares physical and thermal contact from the specimens in the optical cavity. This accessory enables the FCS2 with its perusable, laminar flow optical cavity and user definable flow characteristics to be used for applications where the specimen temperature needs to be below ambient.

Coolant circulation:

Gravity, by means of a siphon or an electric circulating pump is then used to create a flow of chilled fluid through the heat exchange cavity in the chamber (the space in-between the Microaqueduct Slide and the glass window of the FCS2 Cooling Adapter). The cells remain safely enclosed in a separate optical enclosure. If more thermal transfer is necessary, tubing can be installed into the FCS2 base (CFCS2 Link).  An optional recirculating chiller unit is also available with a temperature range of 0 to 45 degree C.

Note: If you are using high N.A. objectives at below ambient temperatures, it will be necessary to cool the objective and thermally isolate the objective from the scope. The optional Thermal Isolator is necessary so that when cooling the objective, heat is also not pulled through the nosepiece causing further stress on the objective.  It will also prevent condensation from forming on the lower element of the objective because a gas port is provided to purge the adapter with dry air. An anti-reflection coated window optimized for 340-700nm seals the bottom of the adapter.
